Popis v původním jazyce: Occurrence of heavy metals in the environment and their entry into the food chain, with all their negative properties is an ongoing problem. The most important role in solving this problem plays rapid and easy-to-use systems for detection of heavy metals in environment (Cd, Pb, As, Hg). Metallothionein is an intracellular protein which has been known since 1957, and which has very good affinity for heavy metal ions. In its structure up to seven of divalent metal ions can be bound. We attempted to use this feature for designing of our biosensor. Recently there is a greater demand for biosensors where the detection feature is provided via bacteria, peptides, proteins or nucleic acids. The resulting biosensors are characterized by a high specificity and low detection limits. In addition to these properties (the biosensor can be modified to be applicable in situ) but with similar or almost similar results as laboratory measurements. The main objective of this study is to design a biosensor which would selectively detect heavy metals in the analyzed samples contaminated by Cd(II) and/or Zn(II). Construction of the biosensor was as follows: the surface of the hanging mercury drop electrode (HMDE) has been modified by the metallothionein (1 uM, 120 s), which due to its characteristics can bind heavy metals. This biosensors was consequently used for detection of cadmium(II) and zinc(II) ions in the presence of NaCl (0.2 M, pH 6.4) as supporting electrolyte and under the following experimental conditions: modulation time 0.057 s, time interval 0.2 s, start potential -1.2 V, end potential -0.3 V, scan rate 5.25 mV/s and modulation amplitude 25 mV using DPV. In cases of both metal ions, we detected a change in base signal (DPV measurement of metallothionein modified HMDE). We found that if we measured zinc the signal corresponding to ZnMT (potential between -1.1 to -1.0 V) complex enhanced with increasing zinc concentration.
